Voltage-gated sodium channel Nav1.7, Nav1.8 and Nav1.9 play important roles in the pain pathway such as in dorsal root ganglion neurons, they have been hot targets in recent years in the pain drug discovery and development field. Nav1.7, in particular, has been proven to be involved in the pain mechanism. Selective potent Nav1.7 inhibitors could alleviate the pain while avoiding many side effects of current pain drugs.
PharmaCore Labs provides manual patch clamp human Nav1.7 (hNav1.7) assay to evaluate drug's inhibition effect on hNav1.7 channel.
In vitro electrophysiology hNav1.7 assay with manual patch clamp:
Assay platform/method | Conventional patch clamp |
Cells used in the assay | CHO hNav1.7 stable cell line |
Measured parameters | Whole cell current of hNav1.7 channels |
Test concentrations | 5 doses for IC50 |
Data point repeats | N ≥ 3 for each concentration |
Note: Positive control: 1 μM TTX or 10 μM Amitriptyline is used in the assay to ensure cell quality and normal response.
